男性不育精子染色质结构与尿酸和丙二醛含量关系研究

摘    要:目的:探讨不育精液中活性、精子染色质结构、MDA与尿酸之间的关系。方法:依据WHO诊断标准,选择不育者43例,生育者30例,进行常规精液分析;丙二醛(MDA)测定用TBA显色法;尿酸含量测定采用尿酸酶-过氧化物酶偶联法,将精子标记相应的荧光染料,用流式细胞仪检测精子的染色质状态。结果:生育组精子活率、密度、UA浓度明显高于不育组(P<0.01),精子畸形率、MDA浓度、DNA碎片率明显低于不育组(P<0.01)。且DNA碎片率与精子畸形率呈正相关(r=0.371,P<0.01),与精子活率呈负相关(r=-0.349,P<0.01)。结论:不育患者精液中脂质过氧化(LPO)反应启动,抗氧化物尿酸含量下降,使精子DNA中毒受损,畸形率升高,密度和活率下降;从常规、生化指标说明精子DNA受损的情况,为基层临床诊治男性不育症提供更有力的证据和更好的帮助。

Investigation on relationship among chromatin structure of sperms, uric acid and malondialdehyde contents in men with infertility
Abstract:Objective: To explore the relationship among activity of sperms,chromatin structure of sperms,malondialdehyde ( MDA) and uric acid in men with infertility. Methods: According to WHO diagnostic criteria,43 infertile cases and 30 normal cases were selected,then routine semen analysis was conducted; MDA level was detected by TBA method; uric acid level was detected by uricase peroxidase coupling method; the status of chromatin was detected by flow cytometry after labeling sperms with corresponding fluorescent dye. Results: The survival rate,density and UA concentration of sperms in fertile group were significantly higher than those in infertile group ( P <0. 01) ; the rate of sperm deformity,MDA concentration and DNA fragment percentage in fertile group were significantly lower than those in infertile group ( P < 0. 01) ; there was a positive correlation between DNA fragment percentage and the rate of sperm deformity ( γ = 0. 371,P < 0. 01) ,and there was a negative correlation between DNA fragment percentage and survival rate of sperms ( γ = 0. 349,P < 0. 05) . Conclusion: Start of lipid peroxidation reaction in semen of infertile patients and decrease of uric acid content induce poisoning and impairment of DNA in sperms,increase of sperm deformity rate,decrease of density and survival rate; demonstrating the impaired status of DNA in sperms from conventional and biochemical indicators may provide more powerful evidences and helps for diagnosis and treatment of male infertility in primary hospitals.
