人参皂苷白蛋白纳米微球对人肺腺癌A549细胞生长抑制作用研究

摘    要:目的探讨载20(s)-人参皂苷白蛋白纳米微球(SPG-Rg3-HAS-NP)对人肺腺癌A549细胞体外增殖抑制作用。方法以人肺腺癌A549细胞为研究对象,分为A、B、C、D 4个给药组。A组为生理盐水对照组,B组为空白白蛋白纳米微球组,C组为20(s)-人参皂苷Rg3(SPG-Rg3)组,D组为载SPG-Rg3-HAS-NP组。采用MTT法观察给予不同浓度药物后24 h、48 h、72 h和96 h 4个时间点人肺腺癌A549细胞生长抑制率。结果 A组与B组对人肺腺癌A549细胞无细胞毒作用。C组与D组对人肺腺癌A549细胞均具有显著的细胞生长抑制作用,且给药浓度和时间呈一定的浓度依赖性和时间依赖性。C组在80μg/mL浓度以后进入平台期的趋势,而D组的细胞生长曲线仍继续呈上升的趋势,72 h后于20μg/mL浓度处抑制率已超过C组(P〈0.05)。结论 SPG-Rg3-HAS-NP具有的缓释作用,对人肺腺癌细胞增殖具有显著的体外细胞生长抑制作用,呈浓度依赖性和时间依赖性。

Abstract:Objective To investigate the load 20 (s) - ginsenoside albumin nanoparticles ( SPG - Rg3 - HAS - NP) inhibitory effect on the proliferation of human lung carcinoma cell A549 in vitro. Methods The human lung adenocarcinoma cell line A549 was regarded as the research object, and they were divided into A, B, C and D groups: A group was normal saline control group, B group was blank albumin nanospileres group, 2 C group was 20 (s) - ginsenoside ( SPG - Rg3 ) group and D group was SPG - Rg3 - HAS - NP group ; Human lung adenocarcinoma 3.549 cell growth rate inhibition was detected after 24h ,48h, 72h and 96h being treated with different drug concentration by MTT assay. Results The group A and the group B had no cytotoxicity on human lung adenocarcinoma A549 cell. The group C and the group D on human lung adenocarcinoma A549 cells growth significantly inhibited the growth of the cells, and human lung adenocarcinoma A549 cells growth had concentration -dependence and time - dependence. The group C cells growth seemed to be platform after concentration of 80μg/ml, but the group D ceils growth had a continue ascend curves. The D group had higher inhibitory effect than the C group after concentration of 20μg/ml SPG - Rg3 and after 72h ( P 〈 0. 05 ). Conclusion SPG - Rg3 - HAS - NP has sustained release effect, has a significant anti - proliferation effect on human lung cancer cells, and has concentration - dependence and time - dependence
