氟化钠与纳米二氧化钛对人支气管上皮细胞的联合作用

摘    要:目的]探讨氟化钠(NaF)与纳米二氧化钛(nano-TiO2)单独及联合染毒对人支气管上皮细胞株(16-HBE)氧化应激及细胞凋亡的作用。方法]对生长状态良好的16-HBE细胞,单独及混合加入NaF(mg/L,F^-)和nano-TiO2(mg/L)染毒,终浓度分别为:(0.0,0.0),(10.0,0.0),(20.0,0.0),(30.0,0.0),(0.0,10.0),(10.0,10.0),(20.0,10.0),(30.0,10.0)。72h后,四甲基偶氮唑盐(MTT)法检测16-HBE的存活率,Hoechst 33258染色观察细胞凋亡,黄嘌呤氧化酶法测定总超氧化物歧化酶(T-SOD),硫代巴比妥酸(TBA)法测定丙二醛(MDA);硝酸还原酶法测定一氧化氮(NO)。结果]30.0mg/LNaF(以F^-计)单独染毒组及各联合染毒组的16-HBE存活率均较对照组降低(P〈0.05)。30.0mg/LNaF(以F^-计)和10.0mg/Lnano-TiO2联合染毒组的T-SOD低于对照组(P〈0.05);各联合染毒组的MDA均高于对照组(P〈0.05);各染毒组NO均高于对照组(P〈0.05).经单因素方差分析,NaF及nano-TiO2联合染毒对MDA和NO有交互作用(P〈0.05)。20.0、30.0mg/LNaF(以F^-计)的单独染氟组及各联合染毒组的16-HBE细胞凋亡率均高于对照组(P〈0.05),未发现NaF和nano-TiO2对16-HBE细胞凋亡存在交互作用。单独染氟或氟联合nano-TiO2染毒:氟与T-SOD呈负相关(P〈0.01);氟与MDA、NO、细胞凋亡率呈正相关(P〈0.05或0.01)。结论]NaF与nano-TiO2联合染毒可能增大对16-HBE的氧化损伤作用;随着氟浓度增高,16-HBE氧化应激水平及细胞凋亡均加重。
