九香止泻片对湿热型泄泻肠易激综合征模型大鼠AQP4、PKA与cAMP表达的影响

摘    要:目的探讨九香止泻片对湿热型泄泻肠易激综合征(IBS)模型大鼠肠道水通道蛋白4(AQP4)、cAMP依赖性蛋白激酶(PKA)与环腺苷酸(cAMP)的影响。方法 40只SD大鼠,随机分为空白组、模型组、九香止泻片组、盐酸洛哌丁胺对照组,每组10只,除空白组外,其余各组大鼠在造模成功后分别给予不同药物或蒸馏水连续灌胃7 d,腹腔麻醉取血处死后取结肠组织测定大鼠肠道AQP4、PKA表达与cAMP含量。结果与空白组比较,模型组肠道AQP4、PKA蛋白表达均显著降低(P<0.05),cAMP含量下降,差异有统计学意义(P<0.05)。与模型组比较,九香止泻片组和盐酸洛哌丁胺对照组肠道AQP4、PKA蛋白表达均显著增强(P<0.05),cAMP含量显著增加(P<0.05);九香止泻片组和盐酸洛哌丁胺对照组比较差异无统计学意义(P>0.05)。结论九香止泻片可能通过激活cAMP-PKA信号通路调控湿热型泄泻IBS模型大鼠肠道中AQP4的表达进而改善腹泻症状,是治疗湿热型泄泻IBS的有效方药。

Abstract:Objective To investigate the effects of Jiuxiang Zhixie tablet (JXZXP) on AQP4, PKA and cAMP expression of irritable bowel syndrome (IBS) rats with damp-heat diarrhea type. Methods 40 SD rats were randomly divided into 4 groups as follows: blank group, model group, JXZXP group and loperamide hydrochloride positive group, 10 rats in each group. Other mice ex- cept for mice in blank group were given different drugs or distilled water respectively after modeled (ig 7 d). After treatment, all mice were killed and taken for colon tissues for observations of AQP4, PKA expression levels and cAMP content. Results Compared with blank control group, the AQP4, PKA expression levels and cAMP contents of colon in model group were reduced significantly(all P〈0.05). Compared with model group, the AQP4, PKA expression levels and cAMP contents of colon in JXZXP group and loperamide hydrochloride group were increased significantly (all P〈 0.05), But the differences between JXZXP group and loperamide hydrochloride group were no sta- tistically significant(P〉0.05). Conclusion JXZXP may be improve the diarrhea symptoms of IBS rats with damp-heat diarrhea type by activating the expression of AQP4 in the intestinal tract of the cAMP-PKA signaling pathway and is an effective medicine.
