Kiss-1基因启动子甲基化与结直肠癌的关系

目的探讨结直肠癌组织中Kiss-1基因启动子甲基化状态与临床病理学特征的关系及临床意义。方法采用甲基化特异性PCR检测142例正常结直肠组织、23例结直肠腺瘤组织、73例结直肠癌组织和相应癌旁组织中Kiss-1基因启动子甲基化状态。结果结直肠癌组织中Kiss-1基因启动子甲基化阳性率为82.2%,癌旁组织为30.1%,结直肠腺瘤组织为21.7%,正常结直肠组织为6.3%,总体比较差异有统计学意义(P<0.05)。Kiss-1基因启动子甲基化阳性率与结直肠癌的分化程度、浸润深度、淋巴结转移及远处转移有关(P<0.05)。结论 Kiss-1基因启动子甲基化可能促进结直肠癌的发生、发展与转移,Kiss-1基因启动子甲基化水平有可能成为评估结直肠癌转移风险及预后的指标之一。

Relationship Between Kiss-1 Gene Promoter Methylation and Colorectal Carcinoma

Objective To research the relationship between Kiss-1 gene promoter methylation and clinicopathological features of colorectal carcinoma and its clinical significance. Methods Kiss-1 gene promoter methylation was detected respectively by methylation-specific PCR in 142 cases of normal colorectal tissues,23 cases of colorectal adenoma tissues,73 cases of colorectal carcinoma tissues and paraneoplastic tissues. Results The positive expression rate of Kiss-1 gene promoter methylation was 82.2% in the colorectal carcinoma tissues,30.1% in the paraneoplastic tissues,21.7% in the colorectal adenoma tissues and 6.3% in the normal colorectal tissues(P<0.05).There were the significant correlation between Kiss-1 gene promoter methylation and the differentiation,depth of invasion,lymph node metastasis and distant metastasis(P<0.05). Conclusion The Kiss-1 gene promoter methylation may contribute to the occurrence,development and metastasis of colorectal carcinoma,Kiss-1 gene promoter methylation may be used as a reference indicator of the risk of colorectal carcinoma metastasis and prognosis.
