活血胶囊对动脉粥样硬化兔平滑肌细胞凋亡及Bcl-2、Bax蛋白表达的影响

目的探讨活血胶囊干预对动脉粥样硬化兔主动脉平滑肌细胞凋亡及凋亡相关因子Bcl-2、Bax蛋白表达的影响。方法通过高脂饮食建立实验性动脉粥样硬化兔动物模型,采用TUNEL法检测平滑肌细胞凋亡,SP免疫组化法检测凋亡相关因子Bcl-2、Bax蛋白表达。结果与模型组比较,活血胶囊低、高剂量组均能降低兔主动脉平滑肌细胞凋亡的凋亡指数(P<0.05,P<0.01),降低Bax蛋白表达(P<0.05,P<0.01),升高Bcl-2蛋白表达(P<0.05,P<0.01),并呈现一定的量效关系(P<0.05)。结论活血胶囊可能通过上调Bcl-2蛋白表达,下调Bax蛋白表达,减少平滑肌细胞凋亡,从而抑制动脉粥样硬化斑块的形成。

Effect of Huoxue Capsule on the apoptosis of vascular smooth muscle cell and expression of Bcl-2 and Bax protein on rabbits with atherosclerosis

Objective To observe the effect of Huoxue Capsule on the apoptosis of vascular smooth muscle cell(VSMC) and expression of Bcl-2 protein and Bax protein on rabbits with atherosclerosis.Methods The experimental model of rabbits with atherosclerosis were induced by atherogenic diet.The cell apoptosis were detected by TUNEL staining,and the expression of Bcl-2 protein and Bax protein were examined by SP immunohistochemical staining.Results Compared with the experimental model of rabbits,treatment with both high dose and low dose of Huoxue Capsule decreased the apoptotic index of VSMC and the expression of Bax protein(P<0.05,P<0.01),and increased the expression of Bcl-2 protein(P<0.05,P<0.01).In addition,the effect of treatment with high dose of Huoxue Capsule was better than with low dose of Huoxue(P<0.05).Conclusion Huoxue Capsule increased expression of Bcl-2 protein and decreased expression of Bax protein in rabbits with arteriosclerosis,and decreased the apoptosis of VSMC,which might be associated with its inhibiting effect on atherosclerotic plaque.
