吗氯贝胺与米帕明治疗抑郁症的对照研究及血药浓度测定

为评价新型单胺氧化酶抑制剂吗氯贝胺的抗抑郁疗效和不良反应,选用米帕明为对照药进行随机对照研究。疗效采用汉密顿抑郁量表和临床印象量表每周评定一次。不良反应采用Asberg's抗抑郁药副作用量表每周评定一次和实验室检查每二周一次。同时测定吗氯贝胺的血药浓度以探讨其与疗效和不良反应的关系。经对两组各18例抑郁症患者为期四周的治疗观察。结果显示,吗氯贝胺的疗效与米帕明相似,抗胆碱能不良反应较米帕明少,对血压也无明显影响。血药浓度与剂量相关,与疗效和不良反应无显著相关。

A Randomized Control Study of Moclobemide and Imipramine in Depression and Plasma Concentration Monitoring of Moclobemide

In order to compare antidepressive efficacy and adverse reaction of moclobemide with that of imipramine in depression. A randomized comparative trial was carried in 36 inpatients fitted CCMD- I -R criterion for depression.After one week placebo washout, a four -week dosage regime of 400 mg/d moclobemide and 200mg/d imipramine were given 18 cases of modobemide group and 18 cases of imipramine group, respectively. The efficacy was rated by Hamilton depression rating scale and clinical global inventory, and the side -effects were assessed by Asherg's rating scale at baseline and weekly during therapy period. Plasma concentrations of moclobemide were measured on 7,14,28 day for investigating the relationship between therapeutic efficacy, and plasma drug level. The results showed that moclobemide was as antidepressive effective as imipramine, and much less anticholinergic side -effects than imipramine. Steady -state plasma moclobemide concentration was correlated with oral dosage and not significantly correlated efficacy and side-effects.
