摘    要:目的 研究白血病患者WT1启动子区域DNA甲基化与WT1表达水平之间的关系及临床意义.方法 利用RT-PCR和甲基化特异性PCR(MSP)检测64例白血病患者、56例对照者骨髓内WT1mRNA水平及DNA甲基化状态.结果 骨髓中WT1表达率与WT1启动子DNA甲基化程度呈负相关性,在急性白血病患者(初诊、复发及未缓解者)和慢粒进入加速期和急变期患者骨髓中WT1启动子DNA甲基化率低于健康者和非白血病患者.结论 WT1启动子区域DNA甲基化抑制WT1mRNA表达.激发WT1启动子区甲基化从而抑制WT1表达,与急性白血病发生转化有关,可以作为治疗和预防白血痛发生的一个研究方向和方法.
