松果体摘除及褪黑激素对大鼠睾丸间质caspase-3和eNOS的影响

摘    要:目的探讨松果体摘除对睾丸间质caspase-3和eNOS表达的影响.方法SD成年雄性大鼠45只分成5组(1)正常对照组,(2)假手术组,(3)松果体摘除组(PX+NS),(4)松果体摘除加低剂量褪黑激素(PX+MT7.5 mg/kg),(5)松果体摘除加高剂量褪黑激素组(PX+MT 15 mg/kg).免疫组织化学SABC法检测caspase-3和eNOS在睾丸间质的免疫活性表达.结果与对照组和假手术组相比,PX+NS组的caspase-3和eNOS的免疫活性显著减少(P<0.01);与PX+NS组相比,PX+MT 7.5 mg/kg组和PX+MT 15 mg/kg组caspase-3和eNOS的免疫活性显著增加(P<0.01);PX+MT 15 mg/kg组与PX+MT 7.5 mg/kg组相比,caspase-3免疫活性无明显变化,eNOS免疫活性明显增加(P<0.05).结论松果体摘除能抑制睾丸间质细胞凋亡,并可被外源性褪黑激素恢复,但与褪黑激素剂量高低无关;褪黑激素能促进睾丸间质eNOS活性表达,且增加MT剂量可进一步增加其活性.

Abstract:Objective To investigate effect of pinealectomy and melatonin on caspase-3 and eNOS in interstitial tissue of rat testis. Methods Forty-five SD mature male rats was divided into five grous, control group, sham group, pinealectomy(PX)+normal saline(NS) group, PX+7.5 mg/kg MT group and PX+15 mg/kg MT group. Immune activity of caspase-3 and eNOS was determined by immunohistochemistry. Results Compared with control group and sham group, the immune activity of caspase-3 and eNOS in PX+NS group decreased significantly(P<0.01). Compared with PX+NS group, the immune activity of caspase-3 and eNOS in PX+MT 7.5 mg/kg group and PX+MT 15 mg/kg group increased significantly(P<0.01). Compared with PX+MT 7.5 mg/kg group, PX+MT 15 mg/kg group showed no significantly different(P>0.05) in the immune activity of caspase-3, but increased obviously the immune activity of eNOS than 7.5 mg/kg group(P<0.05). Conclusion The apoptosis of Leydig cells was inhibited by pinealectomy and was restored by exotic MT, but has no correlation to the dose of MT. The activety of eNOS was promoted by MT giving and with dosage dependently.
