骨巨细胞瘤p73、p15蛋白表达研究

摘    要:目的观察骨巨细胞瘤(GCT)中p73、P15蛋白的表达,探讨其临床意义及相关性。方法30例骨巨细胞瘤标本分别按照Ennecking分期、有无复发、有无转移进行分组。应用S-P免疫组化方法检测标本组中的p73蛋白和P15蛋白的表达。结果p73蛋白和P15蛋白在GCT中的阳性表达率分别为40.0%和56.7%;p73蛋白和P15蛋白的表达强度随Ennecking分期的增高而增加(P〈0.01),二者在复发组的表达均高于无复发组(P〈0.05,P〈0.01);此外,P15蛋白的表达在转移组高于无转移组(P〈0.01);p73、P15蛋白在GCT中的表达呈正相关(P〈0.05)。结论p73、P15蛋白的表达与骨巨细胞瘤的生物学行为有相关性,有助于评估肿瘤的预后。p73和P15的相关性可能是肿瘤抑制性基因网络性结构复杂性的表现。

Abstract:Objective To investigate the expression of p73 and p15 protein in giant cell tumor of bone and their clinical significance. Methods 30 specimen grouped according to the Ennecking staging system and whether recurred or metastasized. p73 and p15 protein from tumor specimen were examined by immunohis- tochemistry S-P technique. Results The positive expression rate of p73 protein and p15 protein in the GCT samples are 40% and 56.7% respectively. The expression of p73 and p15 protein increased along in high stages tumors (P<0.01) and higher in recurred cases t (P<0.05, P<0.01). Expression of p15 protein is also higher in the metastasized cases (P<0.01). Expression of p73 protein and p15 protein were proved to correlate after the Spearman relevance examination (P<0.05). Conclusions Expression of p73 and p15 protein present relevance to the biology behavior of giant cell tumor of bone and may be used for evaluation of clinical prognosis. The correlation of P73 and p15 is possibly the performance of the complex network structure of tumor suppression gene.
