头穴丛刺法联合川芎嗪对大鼠脑梗死后神经元突触重建的影响

摘    要:目的:探析头穴丛刺法联合川芎嗪对大鼠脑梗死后神经元突触重建的影响。方法:行简单随机法将72只SD大鼠分组,备制大脑中动脉闭塞(MCAO)脑梗死模型,A组18只(MCAO模型对照),B组18只(头穴丛刺法),C组18只(川芎嗪),D组18只(头穴丛刺法+川芎嗪)。B组、D组取百会及穴位邻近左侧与右侧2 mm处针刺;C组、D组以盐酸川芎嗪注射液20 mg/(kg·d);A组以同剂量0.9%氯化钠溶液注射。于MCAO建模7、14、28 d比较各组星形胶质细胞、突触个数,检测突触素、GFAP的含量。结果:随时间延长,各组神经元突触、星形胶质细胞个数均有增长,突触素、GFAP表达均有提高,其中B、C、D组增长明显(P<0.05)。14 d、28 d D组神经元突触、星形胶质细胞个数及突触素、GFAP表达均显著高于A、B、C组,差异有统计学意义(P<0.05),其次B、C组14 d、28 d显著高于A组(P<0.05),B、C两组7、14、28 d均无统计学意义(P>0.05)。结论:川芎嗪联合头穴丛刺可促进MCAO脑梗死大鼠神经元突触增殖,增强突触素、GFAP表达,提高突触效能传递,为神经功能重塑提供基础,保护神经元超微结构,修复神经功能。

Abstract:Objective:To investigate the effects of scalp acupuncture combined with ligustrazine on neuron synaptic reconstruction after cerebral infarction in rats.Methods:72 SD rats were randomly divided into groups to prepare a model of middle cerebral artery occlusion(MCAO)cerebral infarction.Group A had 18 rats(MCAO model control),group B had 18 scalp acupuncture methods,C There were 18 rats in the group(ligustrazine)and 18 rats in the group D(head acupoint plexus method+ligustrazine).Acupuncture was performed at Baihui and acupoints 2 mm to the left and right of the points in group B and group D; ligustrazine hydrochloride injection 20 mg/(kg·d)was used in groups C and D; and the same dose of 0.9% chlorine Sodium chloride solution injection.The numbers of astrocytes and synapses in each group were compared on the 7 th,14 th,and 28 d of MCAO modeling,and the contents of synaptophysin and GFAP were detected.Results:Over time,the number of synapses and astrocytes in neurons increased,and the expressions of synaptophysin and GFAP increased in all groups,and the increase in groups B,C,and D was significant(P<0.05); 14 days The number of neuron synapses,astrocytes,and synaptophysin,and GFAP expression in groups D and 28d were significantly higher than those in groups A,B,and C,with significant differences(P<0.05),followed by groups B and C 14 d and 28 d were significantly higher than those in group A(P<0.05),and there were no statistically significant differences between groups B and C on days 7,14,and 28(P> 0.05).Conclusion:Ligustrazine combined with scalp acupuncture can promote neuronal synaptic proliferation,enhance synaptophysin and GFAP expression,improve synaptic efficacy transmission in MCAO cerebral infarction rats,provide a basis for neural function remodeling,and protect the ultrastructure of neurons,repair nerve function.
