高效液相色谱法在测定田银通脉颗粒中皂苷含量中的应用

摘    要:目的建立田银通脉颗粒中三七总皂苷中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量测定方法。方法采用高效液相色谱法测定田银通脉颗粒中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量。色谱柱为Diamon-silTMC18(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.05%磷酸水(B)梯度洗脱(0~8 min,20%A;8~40 min20%~30%A;40~60 min,30%~45%A);流速为1.0 ml/min;柱温为室温;检测波长:203 nm。结果三七皂苷R1在0.212~3.392μg范围内线性关系良好,r=0.999 6,平均回收率为100.21%,RSD=1.71%(n=9);人参皂苷Rg1在0.832~13.312μg范围内线性关系良好,r=0.999 5,平均回收率为100.24%,RSD=1.62%(n=9),人参皂苷Rb1在0.816~13.056μg范围内线性关系良好,r=0.999 7,平均回收率为99.66%,RSD=1.59%(n=9)。结论所建立的高效液相色谱法测定田银通脉颗粒中的三七皂苷R1、人参皂苷Rg、人参皂苷Rb1含量的方法准确、可靠,专属性强,可有效控制田银通脉颗粒的质量。

Abstract:Objective To establish a method for the determination of notoginsecosideR1, ginsenosideRg1 and ginsen- osideRb1 in Tianyin Tongmai granules by high performance liquid chromatography. Methods high performance liquid chromatography was used to determine the content of notoginsecosideR1 , ginsenosideRg1 and ginsenosideRb1 in Tianyin - Tongmai granules. Chromatographic conditions were as follows:DiamonsilTM C18 column(250 mm ×4.6 mm,5 μn) as the stationary phase; acetonitrile (A) - 0.05 % phosphoric acid (B) (0-8 min,20 % A; 8-40 min 20%-30% A ;40-50 min,30% -45 % A) as the gradient mobile phase; the velocity of flow rate : 1.0 ml· min^- l ; the temperature of column: room temperature; detection wavelength: :203 nm. Results The linear ranges of notoginsecosideR1 , ginsenosideRg1 and Rbl fell withinO. 212-3.392 μg (r=0.9995),0.832-13.312μg (r=0. 9995),0.816-13.056 μg (r=0. 999 7), respectively. The average recovery was 100.21% ( RSD = 1.71%, n = 9), 100.24% ( RSD = 1.62 %, n = 9), and 99.66% ( RSD = 1.59%, n = 9) respectively. Conclusions The established method is accurate, reliable and specific, which can effectively control the quality of Tianyin Tongmai granules.
