胆红素对m—DNB所致大鼠肝细胞DNA合成抑制效应影响的初步探讨

摘    要:本研究通过对大鼠肝细胞DNA摄入[3H]-TdR量的检测,主要探讨了m-DNB对大鼠肝细胞DNA合成的抑制效应及胆红素对m-DNB染毒后肝细胞DNA合成的影响。结果表明:m-DNB组计数值/min(cpm)显著低于对照组,并呈明显量-效关系;当肝细胞受损时,低剂量及后加高剂量胆红素组的cpm显著高于m-DNB、空白及胆红素对照组;m-DNB染毒前、中、后三种给胆红素方式cpm值无显著差异。提示胆红素对m-DNB所致DNA合成抑制效应有拮抗作用,可促进肝细胞DNA的合成,从而对肝细胞再生有促进作用。

Abstract:The uptake of TdR was used to observe the inhibitory effects of m DNB on the DNA synthesis in the rat hepatocytes and the protective effects of bilirubin.The results showed that the cpm value in experimental groups were significantly lower than that of control in a dose response relationship manner,in case of liver injury,the cpm value of bilirubin groups,both in lower dose and posttreated higher dose were higher significantly than that in those groups,as m DNB ,control and bilirubin control.The order of bilirubin treatment,before or after m DNB intoxication had no effect on the values of cpm.The study implied that bilirubin would have the antagonist effects on the DNA synthesis in rat hepatocytes exposed to m DNB and promote the DNA synthesis and hepatocyte regene ration.
