骨髓基质细胞对脑创伤大鼠神经营养因子表达水平的影响

摘    要:目的:研究骨髓基质细胞(BMSCs)治疗脑创伤大鼠后内源性的神经生长因子(NGF)和脑源性神经营养因子(BDNF)的影响。方法:采用大鼠骨髓中提取出来的骨髓基质细胞,进行体外扩增后,静脉移植于大鼠脑创伤模型中,在治疗后1、3、5、7、14d,ELISA法检测损伤半球NGF和BDNF的浓度,并与对照组对比,进行统计学分析。免疫组化法检测骨髓基质细胞在损伤脑组织中的表达。结果:骨髓基质细胞经股静脉移植后可在损伤的脑组织中表达。治疗组NGF和BDNF的表达于伤后1d至7d逐渐增加,于第7天达到高峰,至第14天降至较低水平。结论:骨髓基质细胞经股静脉移植后可在损伤的脑组织中表达。移植后的骨髓基质细胞可增加损伤脑组织中的NGF和BDNF的表达,与时间有相关性。体外扩增的骨髓基质细胞对于创伤性脑损伤具有治疗作用。

Abstract:Objective:To investigate the effects of marrow stroma cells(MSCs) on the expression of nerve growth factor(NGF) and brain-derived neurotrophic factor(BDNF) after traumatic rat brain injury.Method:Intravenous injection of the MSCs into the rats after their traumatic brain injury.The level of growth factors was measured quantitatively by using enzyme-linked immunosorbent assay(ELISA).Growth factors studied included NGF and BDNF.Result:The MSCs preferentially migrated into the injured hemisphere.NGF and BDNF were expressed in the injured brain of both treated as well as control rats;however,quantitative ELISA studies showed that expression of NGF and BDNF was significantly increased(P<0.05) in the treated group,and correlated with the time.Conclusion:Intravenous administration of MSCs after traumatic brain injury increases the expression of growth factors(NGF,BDNF),which possibly improves the recovery of these rats.
