空间记忆维持期小鼠海马酪氨酸激酶B含量的变化

摘    要:目的观察Morris水迷宫实验中小鼠记忆维持阶段海马酪氨酸激酶B(trkB)蛋白和mRNA的表达情况。方法将28只C57BL/6品系小鼠分为实验组(n=14)和对照组(n=14),实验组参加水迷宫实验,包括4 d训练和1 d探索实验;对照组仅参加1 d探索实验。记录训练期小鼠的潜伏期、游泳距离和平均游泳速度;探索实验中记录小鼠穿越平台次数和平台象限滞留时间百分比。探索实验结束后采用ELISA法测定小鼠海马trkB蛋白含量,采用Real-Time PCR法测定trkB mRNA表达。结果水迷宫训练期,实验组小鼠潜伏期和游泳距离随训练次数增加逐渐缩短(P<0.05);探索实验中实验组小鼠穿越平台次数和平台象限滞留时间百分比明显高于对照组(P<0.05);两组小鼠海马trkB蛋白和mRNA表达差异均无统计学意义(P>0.05)。结论 Morris水迷宫实验显示,小鼠形成对平台空间位置的长期记忆;但在记忆维持阶段,小鼠海马trkB合成没有增加。
