福州汉族献血者RHD和RHCE基因的多态性分析

目的：探讨福州市汉族献血者RHD和RHCE基因的多态性。方法：应用PCR-SSP等技术分析206名福州市汉族献血者的RHD和RHCE基因。用Arlequin3.5软件计算RHD、RHCE基因频率,检验Hardy-Weinberg平衡,计算最大可能性RH单倍型频率。结果：206名福州市汉族献血者都携带RhD抗原,都存在所检测的RHD基因第1、3、4、5、6、7、9和10外显子。206名献血者中,RHD、RHd、RHDel、RHCe、RHcE、RHce和RHCE基因频率分别为0.9660、0.0267、0.0073、0.6788、0.1667、0.0857和0.0688;DCe、DcE、Dce、DCE、dCe、dce、DelCe和DelcE单倍型频率分别为0.6580、0.1661、0.0733、0.0685、0.0145、0.0122、0.0065和0.0008。福州市汉族献血者RHD、RHEC各基因座基因频率均符合Hardy-Weinberg平衡定律（P〉0.05）。结论：福州市汉族人群RHD、RHd、RHCe、RHcE、RHce和RHCE等基因和DCe、DcE、Dce、DCE、dCe、dce等单倍型都达到多态水平。在RhD血型阳性中国人中,可能有相当数量的人携带诸如RHd、RHD1227A、RHD207A、RHD710delC等基因。

The gene polymorphisms of RHD and RHCE loci in Han population in Fuzhou

Objective：To study gene polymorphisms of RHD and RHCE loci in Han population in Fuzhou.Method：The genes of RHD and RHCE loci in 206 blood donors of Han population in Fuzhou were studied with PCR-SSP and data were analyzed by Arlequin 3.5 software.Result：It was carried by 206 donors that the RhD antigen and exon 1,3,4,5,6,7,9,and 10 of RHD gene what were detected.The gene frequencies of RHD,RHd,RHDel,RHCe,RHcE,RHce and RHCE were 0.9660,0.0267,0.0073,0.6788,0.1667,0.0857,and 0.0688 in Han population in Fuzhou,respectively.The haplotype frequencies of DCe,DcE,Dce,DCE,dCe,dce,DelCe and DelcE were 0.6580,0.1661,0.0733,0.0685,0.0145,0.0122,0.0065,and 0.0008 in Han population in Fuzhou,respectively.The gene frequencies of both RHD and RHEC loci were in Hardy-Weinberg equilibrium（P 0.05） in Han population in Fuzhou.Conclusion：The genes of RHD,RHd,RHCe,RHcE,Rhce,and RHCE and haplotypes of DCe,DcE,Dce,DCE,dCe,and dce were polymorphic.There may be a certain number of people carrying RHd,RHD 1227A,RHD 207A,or RHD 710delC gene in RhD blood group-positive people in China.
