摘    要:目的:采用psbA-trnH序列对竹茹、天竺黄及其近缘物种进行DNA条形码鉴定研究,保障临床用药准确、安全。方法:收集竹茹、天竺黄及其近缘物种共71份材料,提取基因组DNA、PCR扩增并双向测序,经CodonCode Aligner V 4.2拼接获得序列,应用MEGA5.0比对分析,计算种内及种间Kimura-2-Parameter(K2P)遗传距离并构建系统发育树。结果:竹茹、天竺黄各基原及其近缘物种基于psbA-trnH序列的种内最大K2P遗传距离均小于其与近缘物种的种间最小K2P遗传距离;NJ树图结合MEGA比对结果显示,基原植物青杆竹与青皮竹之间无法区分,而竹茹、天竺黄及其他基原物种之间均可较好地区分并呈现明显的单系性。结论:基于psbA-trnH序列可以准确地鉴定多基原药材竹茹、天竺黄及其近缘物种。

Abstract:In this study, the psbA -trnH sequence was used as DNA barcoding to evaluate the accuracy and stability for identification of Bamboo shavings, Concretio Silicea Bambusae and their closely related species. 56 samples were collected and conducted the DNA extraction. Obtained sequences were assembled using the CodonCode Aligner V4.2, the genetic distances and NJ tree were computed and constructed using MEGA 5.0. The results shows that the maximum intra -specific K2P distances were less than the minimum inter -specific K2P distances. The NJ tree indicated that Bamboo shavings, Concretio Silicea Bambusae and their closely related species can be distinguished from each other clearly, except for Bambusa tuldoides Munro and Bambusa textilis, which perhaps attributed to their closely genetic relationship.
