摘 要: | 目的:建立以反相高效液相色谱法测定茶色素中没食子酸和咖啡因含量的方法。方法:色谱柱为Novapak ODS分析柱(250mm×4.6mm,5.4μm),流动相为乙酸-甲醇-N,N-二甲基甲酰胺-水(2:3:35:160),检测波长为280nm,流速为1.0mL.min-1。结果:没食子酸、咖啡因的检测浓度分别在8.16~81.60、2.84~28.40μg.mL-1范围内与各自峰面积积分值呈良好线性关系;没食子酸的平均回收率为99.8%(RSD=0.5%,n=6),咖啡因的平均回收率为99.9%(RSD=0.6%,n=6)。结论:本方法快速、灵敏、准确,可用于茶色素的质量控制。

Determination of Gallic Acid and Caffeine in Tea Pigment by RP - HPLC |

Abstract: | OBJECTIVE:To establish an RP-HPLC method for the determination of Callic acid and Caffeine in tea pigment.METHODS:The separation of sample performed on Novapak ODS column(250 mm×4.6 mm,5.4 μm).The mobile phase consisted of acetic acid-methanol-DMF-H2O(2∶3∶35∶160) with a flow rate of 1.0 mL·min-1 and detection wavelength of 280 nm.RESULTS:The linear ranges of Callic acid and Caffeine were 8.16~81.60 μg·mL-1 and 2.84~28.40 μg·mL-1.The average recovery rate of Callic acid was 99.8%(RSD=0.5%,n=6) and that of Caffeine was 99.9%(RSD=0.6%,n=6).CONCLUSION:The method is rapid,sensitive and accurate,and suitable for the quality control of tea pigment. |
