摘    要:目的:对可溶性人血管内皮细胞生长抑制因子(vascular endothelial growth inhibitor,VEGI)基因进行克隆和表达,检测表达产物对血管内皮细胞增殖的抑制活性。方法:将可溶性人VEGI基因克隆入载体pUC19后测序;再将正确的VEGI基因亚克隆入表达载体pPROEXHTb,IPTG诱导表达;初步纯化表达产物,检测对新生牛主动脉内皮细胞增殖抑制活性。结果:可溶性人VEG

Abstract:Objective:To clone and express the soluble VEGI gene and to test the recombinant product for its inhibition against proliferation of new born bovine aortic endothelial cells. Methods: The soluble VEGI gene was cloned into pUC19 vector and verifed by automatic DNA sequencing.The correct soluble VEGI gene was then subcloned into pP RO EXHTb vector and expressed by induction with IPTG.The recombinant VEGI was partially purified by Ni NTA Spin chromatography and tested for its inhibition against proliferation of new born bovine aortic endothelial cells.Results: The soluble VEGI showed marked inhibitory effect on proliferation of new born bovine aortic endothelial cells in vitro . Conclusion: VEGI is a novel inhibitor of endothelial cells growth and may be valuable for angiogenesis inhibition based cancer therapy.
