培养的同基因兔角膜缘干细胞移植治疗兔眼表烧伤

目的:探讨培养的同基因兔角膜缘细胞移植治疗眼表疾病的方法和疗效。方法:采用消化培养法,兔角膜缘组织进行体外培养,载体为冰冻保存人羊膜,培养的细胞形成单层后移植到已去除病变组织的碱烧伤兔眼球表面,6例兔三度碱烧伤眼进行了培养的同基因兔角膜缘干细胞移植,通过HE染色和免疫荧光染色法,观察眼表结构的愈合情况。结果:术后上皮化进程加快(2～3wk,P<0.05),2mo内均形成较稳定的眼表上皮结构,角膜轻度血管化和结膜化。结论:培养的同基因角膜缘干细胞移植术可作为治疗眼表烧伤、重建眼表的有效方法。

Homologous genic epithelial transplantated limbal stem cells in treating alkaliburned ocular surface disease in rabbits

AIM:To explore the method and to evaluate the therapeutic effect of cultured homologous genic corneal limbal stem cells on ocular surface burning disease in rabbits.METHODS:Rabbit limbal tissue was cultured on the glycerin-deserved amnion membrane in vitro by digestion cultural method.The cultured monolayer limbal stem cells with amnion were transplanted onto the rabbit alkali-burned model eyes.HE stain and immunofluorescence stain were used to observe the healing process of operated rabbit ocular surface.RESULTS:Six eyes received cultured limbal stem cells transpaplantion.The healing course of epitheliazation were speeded up postoperatively.The stable ocular surface was established in 2 months.CONCLUSION:The transplantation of cultural homologous genic corneal limbal stem cells can be used as an effective method to treat alkali-burned ocular surface disease.
