系统性红斑狼疮患者外周血干细胞的研究

摘    要:应用甲基纤维素体外培养法对25例系统性红斑狼疮(SLE)患者进行外周血红系造血干细胞(BFU—E)及粒单系造血干细胞(CFU一GM)测定.结果显示活动期患者两者均显著低于正常人(P<0.01,P<0.01)及非活动期患者(P<0.01,P<0.05).去除患者外周血中的单核细胞及T淋巴细胞并不能使之恢复正常.BFU—E及(CFU一GM的降低与血清IgG的浓度呈负相关(r=-0.58,P<0.01及r=-0.49,P<0.01),与抗双链DNA抗体也呈负相关(r=-0.65,P<0.01及r=-0.57,P<0.01).而患者外周血白细胞计数则与CFU-GM呈正相关(r=0.61,P<0.01).提示患者血清IgG的异常增高可导致患者红系、粒单系造血干细胞的减少.它们的减少是SLE活动有价值的指标.

Abstract:In order to detect the peripheral burst-forming units of erythrocyte(BFU-E) and colony-forming units ofgranulocyte/macrophage(CFU-GM) from patients with SUE and explore their clinical significance.Methods: Methylcellu-lose culture was used to assay the colony formation of BFU-E and CPU-GM in 25 patients with SLE. Results:The numberof BFU-E and CFU-GM in active SLE was much lower than that of normal control and inactive patients. Correlative analysis between the number of progenitor cells(BFU-E and CFU-GM) and the concentration of serum IgG was negative (r=-0.58,P<0.01 and r=-0.49, P<0.01). A close inverse correlation between the number of progenitor cells and thelevels of anti-DNA antibodies was also observed(r=-0.65,P<0.01 and r=-0.57, P< 0.01) .Other significant positive relationship was foud between the number of white blood cell and the number of CFU-GM. Conclusion: A decrease ofnumber of BFU-E and CPU-GM may become a useful marker for SLE activity and the growth of BFU-E and CFU-GMcould be suppressed by IgG in the serum of SLE patients.
