肺岩宁颗粒对Lewis肺癌细胞周期G1/S检测点信号通路中MDM2-P53生物轴的影响

摘    要:目的:观察肺岩宁颗粒对Lewis肺癌小鼠细胞周期G1/S检测点信号通路中MDM2-P53生物轴的影响。方法:采用C57BL/6小鼠,造模并随机分为6组:模型组、顺铂组、肺岩宁煎剂组、肺岩宁颗粒低剂量组、肺岩宁颗粒中剂量组、肺岩宁颗粒高剂量组,进行相应药物干预。观察各组抑瘤率、瘤重、体质量、去瘤体质量,流式细胞术检测细胞周期时相分布,RT-PCR测定肿瘤组织MDM2、P53的mRNA表达。结果:肺岩宁颗粒中剂量组瘤重显著低于模型组,差异有统计学意义(P<0.01),与顺铂组比较差异无统计学意义。肺岩宁颗粒中剂量组体质量与模型组比较,差异有统计学意义(P<0.05);与顺铂组比较,体质量明显升高,差异有统计学意义(P<0.01);肺岩宁颗粒中剂量组去瘤后体质量显著高于煎剂组、模型组、顺铂组,差异有统计学意义(P<0.05,P<0.01)。流式细胞术发现肺岩宁颗粒中剂量组的G0/G1比例较模型组和顺铂组显著增多,癌细胞增殖指数也明显降低,差异有统计学意义(P<0.01)。RT-PCR显示肺岩宁颗粒中剂组MDM2 mRNA水平明显低于模型组、顺铂组、煎剂组,差异有统计学意义(P<0.01);肺岩宁颗粒中剂组P53 mRNA表达明显高于模型组、顺铂组和煎剂组,差异有统计学意义(P<0.01)。肺岩宁颗粒高剂量组无小鼠存活,考虑与颗粒剂溶解度有关。结论:在Lewis肺癌模型中,调节细胞周期G1/S检测点信号通路MDM2-P53生物轴可能是肺岩宁颗粒抗肺癌增殖的重要途径之一。
