摘    要:目的 克隆表达幽门螺杆菌鞭毛粘附素基因hpaA,筛选Hp疫苗有效抗原成分。方法采用PCR技术从幽门螺杆菌基因组中扩增hpaA基因,克隆至表达载体pET-28a(+)中,IPTG诱导表达重组蛋白HpaA(rHpaA),鉴定rHpaA蛋白的表达形式。结果 pPHA重组菌以包涵体形式表达rH-paA,表达量约占全菌的35%。纯化后的rHpaA免疫家兔获得了高效价的兔抗HpaA抗血清,且该抗血清能够与不同Hp菌株中相应分子量条带发生抗原抗体反应,并能在一定程度上抑制由Hp引起的RBC凝集现象。结论 Hp鞭毛粘附素HpaA在重组大肠杆菌中能够高效表达,且具有良好的免疫反应性。

Abstract:Objective To clone and express adhesin gene hpaA of Helicobacter pylori and select the effective candidate for Hp vaccine. Methods Gene hpaA amplified from Hp genome was cloned to pET- 28a( + ) vector, then the recombinant strain (pPHA/BL21) was induced to express rHpaA by IPTG. Results rHpaA occupied about 35 % in total thallus protein of pPHA recombinant strain. By way of immunizing rabbit with purified rHpaA, a high - liter of rabbit anti - HpaA serum has been gained which could react to the corresponding molecular weight band of different Hp strains and could block the hemagglutination caused by Hp . Conclusions HpaA of Hp could be generously expressed in the recombinant E. coli and retained satisfied immunoreaclive.
