异丙酚减轻大鼠创伤性脑水肿及相关机制的研究

摘    要:目的观察异丙酚对大鼠创伤性脑损伤后脑水肿和水通道蛋白4(AQP4)的影响。方法健康雄性Sprague-Dawley大鼠,体质量230~280 g,随机分为假手术组(n=8只)、生理盐水组(n=8只)、异丙酚组(n=8只)。按Feeney自由落体撞击法建立大鼠创伤性脑损伤(TBI)模型。生理盐水组术后腹腔注射生理盐水(50 mg/kg),异丙酚组术后腹腔注射异丙酚(50 mg/kg)。干/湿法测定大鼠建模后48 h脑损伤区头侧的组织含水量,取出脑损伤区尾侧切片进行HE染色观察神经细胞的形态学变化。采用反转录-聚合酶链反应检测脑损伤区AQP4的表达。结果异丙酚组较生理盐水组水肿明显减轻(P<0.05),固缩神经元数量及神经元缺失相对减少,脑损伤区组织AQP4表达下调(P<0.05)。结论异丙酚可显著减轻脑水肿,并且可能下调与脑水肿密切相关的AQP4的表达。

Abstract:Objective To investigate the effects of propofol on the cerebral edema and the expression of AQP4 after the traumatic brain injury in rat. Methods The healthy male Spragne-Dawley rats weighing 230- 280 g were randomly assigned to 3 groups : Sham operation group ( n = 8 ) ; Saline group ( n = 8 ) ; Propofol group( n = 8). The experimental TBI model was established by bumpiness of free falling bady according to Feeneys. The saline group was given NS (50 mg/kg), injected intraperituneally after operation. The propofol group was given propofol( 50 mg/kg), injected intraperitoneally after operation. The brain water content of the rostral side around the injured site were measured by the methods of wet and dry weight. The morphology of the caudal side were investigated by HE staining. RT-PCR was used to determine the change of AQP-4 ex- pression. Results The degree of cerebral edema and the loss of the neurous alleviated after propofol given. The expression of AQP4 was also regulated down. Conclusion Propofol can alleviate the edema volume, and regulate down the expression of AQP4 related to the cerebral edema.
