摘    要:目的探讨中等剂量四氧嘧啶诱导1型糖尿病动物模型的建立。方法选择150只SD大鼠,分为实验1、2、3、4组和对照组,每组30只。禁食12 h后,对实验1、2、3、4组分别按140、150、160、170 mg/kg的剂量经腹膜腔注射四氧嘧啶,对照组注射等量生理盐水,24 h后测血糖,随后每2周测血糖1次,饲养6个月。结果实验1、2、3、4组注药24 h后成模率分别为23.3%、30%、30%、33.3%,6个月后变为13.3%、26.7%、30%、26.7%;对照组血糖均为正常。结论采用150~160 mg/kg四氧嘧啶可建立低成模率、稳定而长期存活的1型糖尿病动物模型。

Abstract:Objective To induce type 1 diabetes mellitus animal model with middle dose of alloxan.Methods Totally 150 SD rats were selected and divided into 5 groups(n=30),group 1,group 2,group 3,group 4 and control group.group 1 to 4 were given 140,150,160,170mg/kg alloxan by intraperitoneal injection respectively while control group was given equal mount of normal saline after fasting for 12 hours.Their blood glucoses were measured 24 hours later and once every two weeks until the sixth month.Results After injection for 24 hours,model forming rates in group 1 to 4 were 23.3%,30%,30%,33.3% respectively;Six months later,their model forming rates were 13.3%,26.7%,30%,26.7% respectively.Blood glucose concentration in control group was always normal.Conclusion Type 1 diabetes mellitus animal model with low model forming rate as well as stable and long-term effect can be established by intraperitoneal injection of 150~160 mg/kg alloxan.
