摘    要:目的:用蛋白质组学的方法研究壮骨止痛方治疗骨质疏松的作用机理。方法:建立去卵巢大鼠骨质疏松症模型,分为壮骨止痛方组、模型组、假手术组,13周后骨质疏松症模型成功,提取股骨组织蛋白质样品,双向电泳分离,得到各组股骨组织总蛋白质分子解剖图谱,用图像分析软件,分析各组间差异蛋白质点,MAlDI-TOF-MS质谱分析,结合蛋白质生物信息库(Marx science Ltd database),对各蛋白质初步鉴定。结果:鉴定了2个差异蛋白,分别X1为糖原磷酸化酶,X2为线粒体ATP合酶β基。结论:在壮骨止痛方治疗骨质疏松的过程中,X1、X2这两种蛋白发挥着重要的分子调控作用。

Abstract:Objective: To study the mechanism about Zhuanggu Zhitong Formula on OP in proteomics. Methods : The ovariectomized rats osteoporosis model was established.Rats were randomly divided into three groups: control group, OP model group and Zhuanggu Zhitong Formula group.The pathology of bone was examined after 13 weeks.The femur tissue protein samples were extracted.Two dimensional-electrophoresis and mass spectrometry were used.Marx science Ltd database was used to elementarily identify variational proteins. Results : Two proteins were identified as glucogen phosphorylase and ATP synthase subunit beta. Conclusion : These proteins have been demonstrated to be postmenopausal proteins.These results can provide valuable experimental evidences for the research on the molecular mechanism of OP which was responsed to OVX and Zhuanggu Zhitong Formula in bone.
