摘    要:目的 观察糖基化终末产物(AGEs)对体外培养的人腹膜间皮细胞(HPMC)纤维连接蛋白(FN)和结缔组织生长因子(CTGF)表达的影响.方法 分别以不同浓度的AGEs(0、200、600、1 000mg/L)刺激HPMC 48h后,用RT-PCR法检测FN的表达情况.同时将体外培养的HPMC分为正常组(未加任何刺激因素)和AGEs组(终浓度为600mg/L),两组均予刺激HPMC48h后,用RT-PCR及ELISA法检测细胞内FN和CTGF mRNA及蛋白的表达水平.结果 AGEs呈剂量依赖性上调HPMC FN mRNA的水平;AGEs组FN、CTGF mRNA和蛋白表达水平均较正常组明显增加(P<0.05或0.01).结论 AGEs可诱导体外培养HPMC的FN、CTGF基因和蛋白高表达.

Advanced glycation end products increase CTGF and FN expression in cultured human peritoneal mesothelial cells

Abstract:Objective To investigate the effect of advanced glycation end products (AGEs) on expression of connective tissue growth factor (CTGF) and fibronectin (FN) in human peritoneal mesothelial cells(HPMC). Methods HPMC were stimulated with different concentrations of AGEs (0,200,600,1 000 mg/ L) for 48 hours, the expression of FN were detected by RT-PCR.2. HPMC were divided into 2 groups: control and 600mg/L AGEs group.The expression of mRNA for FN and CTGF were measured by RT-PCR,the expression of FN and CTGF protein were detected by ELISA after 48 hours .Result The expression of FN mRNA in human peritoneal mesothelial celIs(HPMC) increased in a dose-dependent manner after induced by AGEs; Compared with control,600mg/L AGEs promoted markedly the expression of mRNA and protein for FN,CTGF. Conclusion AGEs can increase markedly the expression of mRNA and protein for FN,CTGF.
