胎儿与成人卵巢免疫原性的比较

摘    要:目的 :比较胎儿卵巢、成人卵巢免疫原性的差异 ,并观察培养前后胎儿卵巢免疫原性的变化。方法 :对培养前后胎儿卵巢及成人卵巢的冰冻切片用人白细胞抗原 (HLA DR)单克隆抗体进行免疫组织化学染色 ,观察其阳性细胞形态及分布并计数。结果 :胎儿卵巢内HLA DR抗原含量显著少于成人卵巢 (P <0 .0 0 1) ,且培养后胎儿卵巢HLA DR抗原含量显著低于培养前 (P <0 .0 1) ,随培养天数的增加HLA DR抗原含量逐渐减少 (P <0 .0 1)。结论 :胎儿卵巢的免疫原性低于成人卵巢 ,且培养后进一步降低。以培养一段时间的胎儿卵巢作为移植供体 ,可能提高临床卵巢移植的成功率。

Abstract:Aim:To compare the immunogenicity between fetal ovaries and adult ovaries,and to observe the change of immunogenicity of uncultured fetal ovaries and cultured fetal ovaries. Methods: To observe the expression of HLA DR antigens and the distribution of positive cells by immunohistochemistry method. Results:The amount of HLA DR antigens in fetal ovaries was significantly lower than those in adult ovaries( P <0.001). The amount of HLA DR antigen positive cells in fetal ovaries of three cultural groups was significantly lower than the uncultured group( P <0.01). The data demonstrated that the amount of HLA DR antigens decreased progressively following with the increase of the cultural days ( P <0.01). Conclusion: The immunogenicity of fetal ovaries is lower than that of adult ovaries. The immunogenicity of fetal ovaries can be reduced after tissue culture. The immunogenicity of fetal ovaries will continue to reduce following with the lasting of the cultural days. As grafts of ovary transplantation, the cultured fetal ovaries will reduce the rejection reactivity after transplantation and prolong the survival of ovarian grafts.
