摘    要:目的:探讨HgCl2对子宫雌激素受体的影响及其雌激素样作用机制.方法:30只大鼠行双侧去卵巢手术后随机分为阴性对照组(蒸馏水)、低、中、高剂量HgCl2组(0.04 mg/kg、0.20 mg/kg、1.00 mg/kg)和阳性对照组(0.08 mg/kg雌二醇),连续用药3 d后取子宫,采用放射性配基结合分析法和雌激素受体结合试验检测雌激素受体数量.结果:HgCl2各浓度组放射活性与阴性对照组比较,差异无统计学意义(F=2.32,P=0.103 0).HgCl2各浓度组雌激素受体数量与阴性对照组比较,差异无统计学意义(F=0.663,P=0.624),也无相关性(rs=0.236,P=0.345).结论:HgCl2不影响雌二醇与雌激素受体结合,也不影响雌激素受体的数量,可能通过其他途径显示雌激素样作用.

Abstract:Aim: To study the effects of HgCl_2 on estrogen receptor (ER) in rat uterine tissue and mechanism of estrogenic effects of HgCl_2.Methods: A total of 30 rats without ovary were randomly divided into 5 groups:negative control group (distilled water),low-,mid-,and high-dose HgCl_2 groups (0.04 mg/kg,0.20 mg/kg,1.00 mg/kg HgCl_2 ),and positive control group (0.08 mg/kg E_2).Three days later,the uterine was removed.The affinity between E_2 and ER, and the quantity of ER were detected using methods of radioactivity ligand bonding test and competed ER bonding test,respectively.Results: Compared with negative control group,no significant difference was observed of radioactivity among HgCl_2 treatment groups( F= 2.32,P=0.103 0).Compared with negative contorl group,no significant difference was observed of quantity of ER among HgCl_2 treatment groups(F=0.663,P=0.624),and no relationship was observed (r_s=0.236,P= 0.345). Conclusion: HgCl_2 does not compete ER with E_2,or affect the quantity of ER.
