聚维酮对膀胱肿瘤T24细胞内阿霉素浓度及Ki-67表达的影响

摘    要:目的 研究聚维酮(PVP)对膀胱肿瘤T24细胞内不同时点阿霉素浓度及Ki-67表达的影响.方法 采用不同浓度的PVP联合阿霉素处理T24细胞,流式细胞术检测各组(ADM组、ADM+2.5%PVP组、ADM+5%PVP组)不同时点(2、24、48、72h)细胞内阿霉素平均荧光强度,光镜下计数各组不同时点细胞数量,免疫组化方法检测各组细胞Ki-67的表达情况.结果 自48h开始ADM+2.5%PVP组细胞计数较ADM组明显减少,自24h开始ADM+5%PVP组细胞计数较ADM+2.5%PVP组明显减少,差异均有统计学意义(均P<0.05);同一时间点3组间细胞内阿霉素荧光强度随PVP浓度的增高而增高,且自2h开始差异即有统计学意义(P<0.05);Ki-67的表达随PVP浓度的升高而降低,3组间比较差异有统计学意义(P<0.05).结论 PVP能明显延长阿霉素对膀胱肿瘤细胞的作用时间,且具有浓度依赖性特点,能够协同阿霉素下调Ki-67的表达,从而抑制膀胱肿瘤细胞的增殖.

Effects of polyvinylpyrrolidone on intracellular adriamycin concentration and expression of Ki-67 in human bladder carcinoma T24 cells

Abstract:Objective To investigate the effect of polyvinylpyrrolidone (PVP) on intracellular concentrations of adriamycin (ADM) and the expression of Ki-67 in human bladder carcinoma cell line T24. Methods T24 cells were incubated with different concentration of PVP combined with ADM. After 2h, 24h, 48h and 72h of incubation the fluorescence intensities of ADM in T24 cells were examined by flow cytometry, the number of cells was counted through light microscope and the expression of Ki-67 was examined by immunohistochemistry. Results Compared to ADM group the number of T24 cells in ADM+2.5% PVP group were significantly decreased after incubating 48h(P〈 0.05 ); and the number of cells in ADM+5%PVP significantly decreased after 24h compared to ADM+2.5%PVP(P〈 0.05 ).There were significant differences of fluorescence intensity between groups at all time points; and the fluorescence intensity rose and expressions of Ki-67 decreased with increasing PVP concentrations (P〈0.05). Conclusions PVP can increase the intracellular ADM concentrations and down-regulate the expression of Ki-67 in human bladder carcinoma T24 cells.
