摘    要:目的 探讨蛋白激酶C(protein kinase C,PKC)亚型α、δ、θ和ζ在宫颈鳞状上皮组织癌变中的表达及作用.方法 (1)Western blot法检测PKC在12例宫颈鳞癌及癌旁正常宫颈鳞状上皮组织中的表达差异;⑵免疫组化法检测另外43例宫颈鳞癌、31例上皮内瘤变(CIN)、32例癌旁正常宫颈鳞状上皮组织中PKC α、δ、θ和ζ的表达.结果 (1)PKC蛋白仅在宫颈鳞癌组织中表达,癌旁正常宫颈鳞状上皮组织中PKC蛋白未见表达.(2)PKCα、δ、θ和ζ在癌旁正常宫颈鳞状上皮组织、CIN、宫颈鳞癌组织中的阳性表达率逐渐增高.(3)PKCα在中低分化宫颈鳞癌的阳性表达率比高分化宫颈鳞癌组织高(P<0.05).PKC δ在II期宫颈鳞癌组织中的表达较I期高(P<0.05).PKC ζ在无淋巴转移的宫颈鳞癌中的阳性表达率比有淋巴转移的宫颈鳞癌高(P <0.05).结论 PKC在宫颈鳞状上皮组织的癌变过程中起了重要的作用,其亚型α、δ、θ和ζ与宫颈鳞癌的临床病理参数有一定的相关性.

Expression of protein kinase C isoforms in cervical squamous cell carcinoma

Abstract:Objective TO investigate the expression of protein kinase C (PKC) isoforms in cervical squamous cell carcinoma. Methods PKC expression was examined by Western blotting in specimens of cancer and pericancerous normal tissue from 12 patients with cervical squamous cell carcinoma. The expressions of PKC isoforms (α、δ、θ and ζ )were detected by immunohistochemistry in other 43 tissues specimens of cervical squamous cell cancer, 32 specimens of corresponding pericancerous normal tissue and 31 specimens of cervical intraepithelial neoplasia. Results Western blot analysis revealed PKC was positively expressed in cervical squamous cancer, but not detected in normal cervical epithelial tissue. Immunohistochemical analysis demonstrated that the expression of PKC isoforms ( α、δ、θ and ζ)was gradually increasing as the tumor progresses. The expression of PKC α was higher in moderately and poorly differentiated tumors than that in well differentiated tumors (P 〈 0.05). The expression of PKC 8 was lower in stage I than that in stage II (P 〈 0.05). There was a significantly increased expres- sion of PKC ζ in tumors without lymphatic metastasis compared to that with lymphatic metastasis (P 〈 0.05). Conclusion The expression of PKC isoforms in tumor cells was associated with clinicopathological parameters of cervical squamous cell carcinoma.
