长期低剂量性激素替代疗法对绝经后妇女外周苯二氮卓受体的影响

摘    要:目的本实验研究长期低剂量激素替代疗法(HRT)对绝经后妇女体内激素水平及外周型苯二氮卓受体(PBR)的影响。方法筛选北京协和医院医护人员中符合入组条件并接受HRT治疗持续4年以上志愿者83人,同时以年龄为配比条件筛选出99人作为对照。以酶联免疫反应法测血浆雌二醇、孕酮、睾酮含量;以同位素标记法测外周血PBR结合活性。结果与对照组相比,HRT组受试者血浆中雌二醇维持在较高水平,两组之间在各个年龄段都有显著差异(P<0.05),而血浆中孕酮、睾酮水平未见显著差异。各年龄段HRT组PBR水平均高于对照组,并在50~59年龄段两组之间有显著性差异(P<0.01)。受试者血中PBR结合活性与雌激素水平呈明显相关性。结论长期低剂量HRT使绝经后受试者体内雌激素水平维持在一个较高浓度的同时血小板上PBR亦处于较高的水平。

Abstract:Objective This study was designed to study the effects of long-term low dosage hormone replacement therapy on the hormone level and peripheral Benzodiazepine receptors (PBR) of post-menopausal women.Methods 83 women with long-term low dose HRT for over 4 years entered the study, 99 people with age and education matched were enrolled in this study as control. All the subjects were from the personnel of Peking Union Hospital. Plasma estradiol, testosterone and progesterone were analyzed with ELISA. The binding activities of peripheral blood PBR were analyzed with isotope labeling method.Results Compared with the control group, there was a significant increase in the level of estradiol (P<0.05) in the subjects with different ages and no difference in the level of progesterone and testosterone of postmenopausal women using HRT. Additionally, postmenopausal women using HRT displayed a higher binding capacity of PBR. A significant difference was found between the 50~59 age stage and control group (P<0.01). Meanwhile, the binding capacity of PBR was significantly correlated with plasma estradiol level.Conclusion These results suggest that long-term low dose HRT could maintain estradiol and PBR binding at higher levels.
