| 摘 要： | 目的 建立和优化人角质形成细胞蛋白质组研究样品制备方法和双向电泳技术条件.方法 体外无血清培养人角质形成细胞,以裂解液成分、上样量的选择和等电聚焦参数为侧重点,通过比较分析不同条件下电泳图谱,建立最佳双向电泳技术条件.结果 成功建立了人角质形成细胞总蛋白制备方法,优化了人角质形成细胞双相电泳技术,获得了重复性和分辫率都较理想的角质细胞总蛋白质双向电泳图谱.结论 优化后的双相电泳条件为人角质形成细胞蛋白质组学分析奠定基础.

| Abstract: | ObjectiveTo establish and optimize the two-dimensional gel electrophoresis(2-DE) and the preparation of protein samples for the proteome of keratinocytes. MethodsThe keratinocytes were isolated from the human skin and cultured in serum-free media.
