三七总皂苷对大鼠血管内膜增生细胞周期蛋白表达的影响

摘    要:目的探讨三七总皂苷对大鼠球囊导管致血管内膜损伤后血管再狭窄血管平滑肌细胞(VSMC)细胞周期蛋白表达的影响及其作用机制。方法将SD大鼠随机分为假手术组、模型组、PNS组和阿托伐他汀组,以球囊导管损伤主动脉内膜后第2天起灌胃给药,连续14d,末次给药后次日(术后第16天)取胸主动脉损伤段,以免疫组织化学法测定损伤血管内膜cyclin D1、cyclin E的表达。结果与模型组比较,增生内膜中cyclinD1、cyclinE表达显著增强,差异均有统计学意义(P〈0.01)。PNS组cyclinD1和cyclinE表达显著降低,差异有统计学意义(P〈0.05,P〈0.01),阿托伐他汀组cyclin D1表达显著降低,差异有统计学意义(P〈0.01)。结论大鼠主动脉内膜剥脱后第16天出现因血管内膜增生引起的血管狭窄,增生内膜中cyclinD1、cyclinE表达均增强,提示其内膜增生主要是由于VSMC增殖所致。PNS可抑制内膜损伤后的血管狭窄。其作用机制与抑制细胞周期蛋白表达。抑制VSMC过度增殖有关。

Abstract:Objective To investigate the effect and mechanism of Panax notoginseng saponin (PNS) on expression of VSMC CyclinD1 and CyclinE induced by balloon catheter injury in rats. Methods SD Rats were randomly divided into 4 groups as following, sham operation, model, PNS (100 mg/kg)and Atorvatatin (10 mg/kg)groups. Aortic intima injury was induced by balloon catheter, and all rats were treated from 2nd day, and for 14 days continuously, at the 16th day, the express of CyclinD1 and CyclinE were observed with immunochemistry. Results Compared with sham operation group, the expression of CyclinD1 and CyclinE were increased obviously in model group(P〈0.01); compared with model group, the expression of CyclinD1 and CyclinE protein in PNS group decreased (P〈0.05, P〈0.01); compared with model group, the expression of CyclinD1 protein in Atorvatatin group decreased (P〈0.01).Conclusion PNS can inhibit the angiostegnosis after injury, which is related that PNS inhibited the expression of CyclinD1 and CyclinE protein and VSMC proliferation.
