摘    要:目的鼻咽癌是一种以放射治疗为主的头颈肿瘤,但部分鼻咽癌对放疗耐受。为了解鼻咽癌放疗耐受机制,本实验拟初步探讨人高分化鼻咽癌细胞株CNE1放疗后的蛋白质改变。方法利用固相pH梯度二维凝胶电泳,建立CNE1放疗前后总蛋白质2DE图谱,利用MALDI TOF MS及数据库搜索初步分析和鉴定部分放疗前后差异蛋白质点。结果高分子量酸性蛋白质在CNE1放疗后20min高表达,其中大多为热休克蛋白质HSPs(heatshockproteins,HSPs)。结论放射能诱导CNE1的热休克蛋白高表达,它们可能与鼻咽癌放疗耐受有关。

Abstract:Objective Nasopharyngeal carcinoma (NPC) is a common head and neck cancer treated mainly by radiotherapy. But some of the NPC are radiation resistance. To understand the intrinsic mechanisms of NPC radiation resistance, the alterations were analyzed for the protein expression of human well-differentiated NPC cell line CNE1 after radiation, which is relatively resistant to radiotherapy. Methods The total protein 2-DE maps of CNE1 cells before and after radiation were established by Two-dimensional gel electrophoresis (2-DE), and some of the differential expressional proteins were identified by MALDI-TOF-MS and database searching. Results The high molecular weight acid proteins were over-expressed 20 min after radiation, most of which are heat shock proteins (HSPs). Conclusion Radiation can induce over-expression of HSPs in CNE1, and these over-expressed HSPs may be relative to CNE1 irradiation resistance.
