摘    要:目的:采用RNA干涉技术干扰宫颈癌HPV16E6基因转录,通过体内动物实验了解其抑制HPV16E6基因的效率。方法:设计合成针对HPV16E6的siRNA,借脂质体转染宫颈癌CaSki细胞,通过体内实验中建立子宫颈癌荷瘤裸鼠模型,将E6siRNA直接注入动物腹腔或瘤体,通过移植瘤体积、重量的变化,肿瘤切片HPV16E6免疫组化,肿瘤细胞凋亡来了解RNA干涉的效果。结果:体内实验中无论腹腔还是瘤内注射HPV16E6siRNA,均明显抑制肿瘤的生长,HPV16E6蛋白表达明显受抑,肿瘤细胞凋亡增加,重复给药较单次给药效果更好。结论:宫颈癌裸鼠移植瘤实验表明RNA干涉HPV16E6基因的效果具有特异性和高效性。

Abstract:Objective:To investigate in vivo the effect of HPV16 E6 siRNA on cervical cancer cell line CaSki cells.Methods:The specific siRNA of HPV16 E6 was synthesized and transfected into CaSki cells by liposome.Cervical cancer in nude mice were established,and siRNA was injected directly into the nude mice peritoneal cavity or subcutaneous tumor.The efficiency of siRNA was evaluated by tumor volume and weight change,HPV16 E6 protein expression and apoptosis of tumor cells.Results:In vivo,E6 siRNA administration groups showed a dramatic effect in inhibiting tumor growth,suppressing expression of E6 protein,inducing tumor apoptosis as compared with the control group.Direct injection of siRNA into subcutaneous tumor resulted in tumor suppression effect similar to via peritoneal cavity,and with additional injection better results could be achieved in cervical cancer CaSki cells.Conclusion:RNA interference to HPV16 E6 is specific and highly efficient in vivo.
