黄芪对哮喘小鼠CD4^+CD25^+调节性T细胞及IL-4IFN-γIL-10的影响

摘    要:目的探讨黄芪对哮喘小鼠CD4^+CD25^+调节性T细胞及IL-4、IFN-γI、L-10的影响。方法将BALB/c小鼠30只随机分为对照组、哮喘组和黄芪组。以卵蛋白(OVA)致敏激发法制备小鼠哮喘模型。酶联免疫吸附试验(ELISA)法检测支气管肺泡灌洗液(BALF)中IL-4、IFN-γ、IL-10及血清中IL-10的含量;流式细胞术(FCM)、反转录聚合酶链反应(RT-PCR)分别检测小鼠脾脏中CD4+CD25+调节性T细胞数量及FoxP3 mRNA表达情况。结果哮喘组小鼠BALF中IL-4含量明显高于对照组(P〈0.01)而低于黄芪组(P〈0.01)。与对照组相比,哮喘组小鼠BALF中IFN-γI、L-10、血清中IL-10含量及脾脏中CD4+CD25+调节性T细胞数量、FoxP3 mRNA表达水平明显降低(P〈0.01);而黄芪组的上述改变较哮喘组显著增加(P〈0.05)。结论 CD4^+CD25^+调节性T细胞参与了支气管哮喘的发病过程,黄芪可通过上调CD4^+CD25^+调节性T细胞、FoxP3 mRNA的表达及增加IL-10含量减轻哮喘炎症。

Abstract:Objective To study the effect of Astragalus(AST) on CD4^+ CD25^+ regulatory T cells and IL-4、IFN-γ、IL-10 in the mice model of asthma.Methods 30 BALB/c mice were divided into 3 groups:control group,asthma group and AST group.Asthmatic mice models were established by ovalbumin intraperitoneal injection.The levels of IL-4、IFN-γ and IL-10 were measured by ELISA.Numbers of CD4^+ CD25^+ regulatory T cells were examined by flow cytometry;the mRNA expression of FoxP3 was tested by RT-PCR.Results The BALF level of IL-4 in asthma group was higher than Control group(P0.01),and lower than AST group(P0.01).Compared with control group,the BALF levels of IFN-γ、IL-10 and the serum level of IL-10 in asthma group,numbers of CD4^+ CD25^+ regulatory T cells and the mRNA expression of FoxP3 were decreased obviously(P0.01).However,the changes of above-mentioned of AST group were increased significantly compared with asthma group(P0.05).Conclusion Numbers of CD4^+ CD25^+ regulatory T cells in spleen and the level of IL-10 in BALF and serum decreased in asthmatic mice which might contribute to the pathogenesis of asthma.AST can increase the numbers of CD4^+ CD25^+ regulatory T cells,the mRNA expression of FoxP3 and the level of IL-10,and thus provides protective effects against asthma.
