Persephin基因转染对缺氧诱导的神经干细胞凋亡的影响

摘    要:目的:探讨重组腺病毒介导Persephin基因治疗对缺氧状态下对神经干细胞凋亡的作用。方法:体外培养C17.2神经干细胞,建立神经干细胞缺氧模型;将携带人类Persephin基因的重组腺病毒感染C17.2神经干细胞;Western-blotting法分析Persephin蛋白的表达;TUNEL法检测凋亡指数,显微镜观察凋亡小体;流式细胞术测定细胞凋亡率的变化。结果:转染pAdPersephin的C17.2神经干细胞成功表达Persephin蛋白;缺氧后神经干细胞凋亡指数为(25.54±4.30)%,而转染pAdPersephin后的细胞凋亡指数显著减少至(10.04±1.32)%(P<0.01),而转染pAdCMVPersephin Persephin反义寡核脱氧核酸细胞凋亡指数为(24.05±3.05)%,与对照组无显著差异(P>0.05)。结论:腺病毒介导的Persephin基因能高效表达Persephin;外源性Persephin对C17.2神经干细胞具有抗凋亡作用,能够提高C17.2神经干细胞对缺氧的耐受性。

Abstract:Objective: To study the effects of Persephin gene on the apoptosis of neural stem cell after hypoxia in vitro. Methods: C17.2 neural stem cell was cultured and infected by recombinant adenovirus containing Persephin gene, then cultured under hypoxic condition. Persephin expression was detected by Western blotting, apoptotic index was measured by TUNEL, and apoptosis bodies was observed under fluorescence microscope, apoptotic ratio was inspected by flow cytometry. Results: The expression of Persephin in pAd Persephin infected cells was significantly increased, furthermore,Persephin gene inhibited the apoptosis of C17.2 neural stem cell. Conclusions: Recombinant adenovirus containing Persephin gene can transfect C17.2 neural stem cell efficiently and the exogenous Persephin expressed highly in vitro. Persephin gene transfection could reduce the apoptosis of C17.2 neural stem cell.
