扶正散结方逆转Lewis肺癌小鼠Th1/Th2漂移相关性研究

目的:探讨扶正散结方对Lewis肺癌小鼠Th细胞因子免疫生物调控作用。方法:建立Lewis肺癌小鼠模型,随机分为荷瘤模型组、环磷酰胺(CTX)组、中药预防组、综合组,每组15只,灌胃及腹腔注射给药,观察移植瘤生长情况,接种后第12天处死动物,称各组瘤重胸腺重,计算抑瘤率、胸腺指数。免疫组化法观察扶正散结方对IFN-γ、IL-4细胞因子影响。结果:与荷瘤模型组相比,环磷酰胺(CTX)组、中药预防组、综合组抑瘤率分别为85.34%、55.25%、87.85%,瘤重低于对照组,差异有显著性(P<0.01)。与CTX组相比,综合组胸腺指数升高(P=0.02<0.05),中药预防组、综合组IFN-γ含量增加(P<0.05)。结论:扶正散结方可改善CTX对小鼠免疫功能抑制,上调Th1细胞分泌的最特异细胞因子IFN-γ含量,下调Th2细胞分泌的最特异细胞因子IL-4含量,逆转Th1/Th2向Th2漂移,抑制肿瘤免疫逃逸,对中医药多靶点肿瘤的免疫治疗提供新思考。

Related Study of Fuzheng Sanjie Recipe on Reversing Th1/Th2Drift in Mice with Lewis Lung Cancer

Objective: To observe the influences of Fuzheng Sanjie Recipe on the growth and the expression of Th cytokine in mice with Lewis lung cancer(LLC).Methods: Lewis mouse model of lung cancer was established and 60mice were randomly divided into 4groups,including the control group,cisplatin(CTX)group,group of Fuzheng Sanjie Recipe and group of Fuzheng Sanjie Recipe combining CTX(Integration group),each group was 15mice,which were treated with various dosage of Fuzheng Sanjie Recipe.The growth of the transplanted tumor was observed,and the mice were killed on the 12th day for weighing tumor and thymus and calculating tumor inhibition rate and thymus index.Imunohistochemical technique SP method was used to examine the expression of the IFN-γ and IL-4cytokines.Results: In cisplatin(CTX)group,group of Fuzheng Sanjie Recipe and Integration group,the tumor was significantly inhibited,the tumor inhibition rate respectively was 85.34%,55.25%,and 87.85%,and tumor weight was lower than that in the control group(P<0.01).Compared with cisplatin(CTX) group,thymus index increased in Integration group,with a significant difference(P=0.02<0.05).The levels of IFN-γ expression were increased in C and D group(P<0.05).Conclusion: Fuzheng Sanjie Recipe has an inhibitory effect on the growth of transplanted tumor in mice with LLC.The mechanism may be related to that Chinese medicinal can improve the expression of the most specific IFN-γ secreted by Th1cell cytokine and decreases the expression of IL-4secreted by Th2cell the most specific cytokine,then induce apoptosis,and the effect can be promoted after they combining with CTX.
