| 摘 要： | 目的:探讨小分子干扰RNA(siRNA)对人食管鳞癌EC9706细胞中桩蛋白(paxillin)表达的抑制作用.方法:使用paxillin siRNA 转染人食管鳞癌EC9706细胞,以对照 siRNA 转染组和正常 EC9706 细胞组为对照,采用免疫细胞化学SP法、Westem blot和半定量RT-PCR技术分别检测转染前后上述各组细胞中paxillin蛋白和mRNA的表达.结果:转染paxillin siRNA后,免疫细胞化学结果显示,3组细胞中paxillin siRNA转染组中paxillin蛋白阳性表达细胞数最低,组间比较差异具有统计学意义(P<0.05);Western blot及RT-PCR结果显示,与正常EC9706细胞组及对照siRNA转染组相比,paxillin siRNA转染组中的paxillin蛋白及mRNA的表达量明显下调,差异有统计学意义(P<0.05).结论:paxillin siRNA能有效抑制人食管鳞癌EC9706细胞中paxillin基因的表达.
