赤雹根总皂苷对佐剂性关节炎大鼠脾淋巴细胞及腹腔巨噬细胞的影响

摘    要:目的研究赤雹根总皂苷对佐剂性关节炎(AA)大鼠脾T淋巴细胞增殖及腹腔巨噬细胞IL-1β、TNF-α合成的影响。方法以弗氏完全佐剂诱导大鼠佐剂性关节炎,将造模成功的50只大鼠随机分为模型组、赤雹根总皂苷160、80、40mg/(kg·d)组和雷公藤12mg/(kg·d)组,另取10只正常大鼠作为对照组。连续灌胃给药21d,给药后第22天处死大鼠。制备大鼠脾细胞悬液,用刀豆蛋白A(ConA)诱导脾T淋巴细胞增殖,MqT法测定增殖指数;制备大鼠腹腔巨噬细胞悬液,用脂多糖(LPS)诱导IL-1β、TNF-α合成,ELISA法检测IL-1β、TNF-α含量。结果与对照组比较,模型组大鼠脾T淋巴细胞增殖反应增强(P〈0.01),腹腔巨噬细胞IL-1β、TNF-α合成增加(P均〈0.01),赤雹根总皂苷能降低其脾T淋巴细胞增殖反应和腹腔巨噬细胞IL-1β、TNF-α合成(P均〈0.01)。结论赤雹根总皂苷对大鼠AA的治疗作用与抑制脾T淋巴细胞增殖及腹腔巨噬细胞IL-1β、TNF-α合成有关。

Abstract:Objective To study the effect of the total saponins of the Thladiantha Dubia roots (SRTD) on adjuvant arthritis (AA) rat' s spleen T lymphocytes proliferation and the synthesis of interleukin-1β (IL-1β) , tumor necrosis factor alpha (TNF-α) of peritoneal macrophage (PM(I)). Methods Fifty AA rats models induced with freund' s complete adjuvant (FCA) were divided into model group, SRTD 160, 80 and 40 mg/(kg·d) groups, and tripterygium 12 mg/( kg·d) group, while ten normal rats were as control group. After intragastric administration for 21 days, all rats were killed and prepared spleen cell suspension. Spleen T lymphocytes proliferation induced by concanavalin A (ConA) was examined with MTT assay. To prepare PM(I) suspension, the synthesis of IL-1β, TNF-α induced by LPS were estimated by ELISA method. Results Compared with control group, AA rat' s spleen lymphocytes proliferation, the content of IL-1β and TNF-α were higher (P 〈0.01 ) , while SRTD could decrease the spleen lymphocytes proliferation and the content of IL-1β, TNF-α (P 〈0. 01 ). Conclusion The treatment of SRTD on AA rats is closely related with the inhibitory effect on spleen T lymphocytes proliferation and the synthesis of IL-1β, TNF-α of PMΦ.
