摘    要:目的 建立并比较测定伪麻黄碱血药浓度的HPLC法、柱前衍生-HPLC法和LC/MS/MS法,为体内低浓度伪麻黄碱的分析提供更为简便、准确的方法.方法 摸索并优化血浆前处理条件、衍生化条件、色谱条件、质谱条件等.结果 3种方法均具有较好的专属性、线性、精密度、回收率和稳定性,可准确、灵敏地检测生物样本中的伪麻黄碱.3种方...

Abstract:OBJECTIVE Establish and compare the methods of HPLC,precolumn derivatization HPLC and LC/MS/MS for determination of pseudoephdrine in plasma in order to provide more simple and accurate methods for the analysis of low-concentration pseudoephdrine in vivo.METHODS The conditions of plasma pretreatment,derivatization,chromatography,MS and so on were explored and optimized.RESULTS Three methods have good specificity,linearity,precision,recovery and stability.They can be used to detect pseudoephedrine in biological samples accurately and sensitively.No significant differences but a satisfactory correlation was found in the results of the three methods.However,compared with the other methods,the average plasma concentration obtained from HPLC was increased a little.CONCLUSION The results of HPLC were a little higher,so precolumn derivatization HPLC and LC/MS/MS were more accurate and reliable to be used in the research of pseudoephdrine in vivo.
