Survivin靶向siRNA抑制结直肠癌细胞增殖作用的研究

目的分析Survivin靶向siRNA抑制结直肠癌细胞增殖的作用。方法将结直肠癌Lovo细胞株进行培养及转染,采用Western blot法、RT-PCR法对Survivin的表达进行检测,应用MTT法对siRNA抑制细胞增殖的作用进行检测。结果 Survivin靶向小干扰RNA抑制结直肠癌SW480细胞株mRNA的构成比为37.1%,抑制结直肠癌SW480细胞株蛋白表达的构成比为45.3%,12 h对结直肠癌SW480细胞的抑制率为23.8%,24 h对结直肠癌SW480细胞的抑制率为30.2%,36 h对结直肠癌SW480细胞的抑制率为35.2%,48 h对结直肠癌SW480细胞的抑制率为38.5%,72 h对结直肠癌SW480细胞的抑制率为39.6%,均显著高于阴性对照组(P均<0.05)。结论 Survivin靶向siRNA抑制结直肠癌细胞增殖的作用显著。

Study on inhibition of survivin targeted on siRNA in colorectal cancer cell proliferation

Objective It is to analyze the effects of survivin targeted on siRNA in inhibiting colorectal cancer cell proliferation.Methods The colorectal cancer Lovo cell was cultured and transfected,the expression of survivin was detected by Western blot method and RT-PCR method,and the inhibition of siRNA to cell proliferation was detected by MTT method.Results Survivin targeted on RNA of SW480 cell for inhibiting colorectal cancer cell mRNA,its composition rate was 37.1%.The composition rate for inhibiting the protein expression of colorectal cancer SW480 was 45.3%.The composition rate for inhibiting colorectal cancer SW480 was 23.8% in 12 hours,The composition rate for inhibiting colorectal cancer SW480 was 30.2% in 24 hours,the composition rate for inhibiting colorectal cancer SW480 was 35.2% in 36 hours,the composition rate for inhibiting colorectal cancer SW480 was 38.5% in 48 hours,the composition rate for inhibiting colorectal cancer SW480 was 39.6% in 72 hours,they were significanty higher than the negative control group(all P<0.05).Conclusion The effect of survivin targeted on siRNA for inhibiting colorectal cancer cell proliferation was significant.
