摘    要:目的 探讨氯喹提高胰岛素敏感性的作用机理。方法 在高脂肪饮食喂养法复制的胰岛素抵抗(IR)大鼠模型中,观察大鼠的胰岛素敏感性的变化以及氯喹对其影响。结果 IR大鼠与正常大鼠相比,骨骼肌2-脱氧葡萄糖(2-DG)摄取值和正常4血糖钳夹技术测定的葡萄糖输注率(GIR)显著下降;肝脏胰岛素降解活性(LIA),空腹血浆胰岛素(FINS)及血脂则显著升高。而在高脂肪饮食喂同时服用氯喹的大鼠与IR大鼠相比,C

Abstract:OBJECTIVE: To investigate the mechanism of chloroquine improving insulin sensitivity. METHODS: The changes in insulin sensitivity and the effects of chloroquine were observed in insulin-resistant (IR) rats induced by high-fat feeding. RESULTS: In IR rats, the glucose infusion rate (GIR) determined by euglycemic clamp and 2-deoxyglucose (DG) uptake in the skeletal muscle were lower than those in normal rats, but the activity of insulin degradation of liver (LIA), fasting plasma insulin (FINS) and plasma lipids were higher. In the high-fat fed rats with chloroquine administration, GIR and 2-DG uptake in the skeletal muscle were increased, and LIA, FINS and some plasma lipid indices were decreased compared with the IR rats. CONCLUSION: Chloroquine can improve the insulin sensitivity and hamper the onset and development of insulin resistance.
