新维甲类化合物5-4A抗癌细胞侵袭作用及其机理研究

摘    要:目的 为探讨新维甲类化合物54 A 对癌细胞侵袭能力的影响及其作用机理。方法利用体外重组基底膜侵袭试验,观察癌细胞的侵袭能力;应用生物化学及形态学方法,检查细胞的恶性表型。结果 54 A 在4 ×10 - 5 mol/ L浓度时,对黑色素瘤高转移株 B16 B L6 细胞膜侵袭具有显著的抑制作用,抑制率为339 % 。结论 54 A 具有抗 B16 B L6 膜侵袭作用。其作用与其抑制肿瘤细胞的运动能力、Ⅳ型胶原酶活力以及改变肿瘤细胞的恶性表型有关。

Abstract:OBJECTIVE: To investigate the effect of 5-4A on the invasion of B16BL6 and to explore its mechanisms of action. METHODS: The invasion of B16BL6 was studied using the reconstituted basement membrane invasion assay. RESULTS: The phenotype of B16BL6 cells was obviously suppressed by 5-4A at the concentration of 4 x 10(-5) mol/L, and the inhibitory rate was 33.9%. CONCLUSION: 5-4A can inhibit invasion of B16BL6 cells and its mechanisms of action might be associated with the suppression of movement of tumor cells, decrease in the expression of type IV collagenase, and reversion of the phenotype of B16BL6 cells.
