大承气汤对内毒素血症小鼠血浆内皮素-1及肾功能的影响

摘    要:目的观察中药大承气汤对内毒素血症血浆内皮素-1及肾功能的影响。方法66只小鼠均分为三组:对照组,内毒素血症组(腹腔注射内毒素,立即、4h用生理盐水灌胃),和治疗组(腹腔注射内毒素,立即、4h用大承气汤灌胃)。注射4h、8h后眼眶取血测定血清血浆内皮素-1(ET-1)和血清肌酐。结果注射内毒素后4和6h,治疗组血浆内皮素一1水平分别为(363.669±31.747)pg/ml和(323.320±43.327)pg/ml,均显著低于内毒素血症组[(470.028±49.430)和(462.917±20.13)pg/ml,均P〈0.01]。注射内毒素后4和8h,治疗组血清肌酐水平分别为(40.1±3.71)μmol/L和(39.29±3.71)μmol/L,均显著低于内毒素血症组[(48.18±5.15)和(47.38±4.14)μmol/L,均P〈0.01]。结论大承气汤能降低内毒素血症血浆内皮素水平和血清肌酐水平,减轻肾脏损伤,其机制可能与抑制内皮素有关。

Effects of Dachengqi Decoction on plasma endothelin-1 and serum creatinin in endotoxemia: experiment with mice

Abstract:Objective To investigate the effects of Dachengqi Decoction (DD) on plasma endothelin-1 and renal function in endotoxemia. Methods Sixty-six mice were randomly divided into 3 equal groups: lipopolysaccharide (LPS) group [undergoing intraperitoneal injection of LPS and gastic perfusion of normal saline (NS) immediate and 4 h after], DD treatment group (undergoing intraperitoneal injection of LPS and gastic perfusion of DD immediate and 4 h after), and control group (undergoing intraperitoneal injection of NS). Blood samples were collected from the eye sockets 4 and 8 hours after the introperitoneal injection to test the plasma endothelin-1 (ET-1) and serum creatinin. Results The plasma ET-1 levels 4 and 8 hours later of the DD group were (363.669±31.747) and (323.320+±43.327) pg/ml respectively, both significantl7 lower than those of the endotoxemia group [(470.028±49.430)and(462.917±20.13) pg/ml respectively, both P〈0.01 ]. The serum creatinin levels 4 and 8 hours later of the DD group were (40.1±3.71) and (39.29±3.71) μmol/L, both significantly lower than those of the endotoxemia group [ (48.18±5.15) and(47.38±4.14) μmol/L respectively, both P〈0.01]. Conclusion DD decreases the levels of plasma ET-1 and serum creatinin, thus reducing the renal injury with the probable mechanism of inhibiting ET- 1 level.
