细胞外信号调节激酶在非小细胞肺癌中的表达及其意义

摘    要:目的 :探讨细胞外信号调节激酶ERK 1和ERK 2蛋白在非小细胞肺癌组织中的表达及其意义。方法 :用固定化蛋白质印迹法检测 5 2例非小细胞肺癌及癌旁正常肺组织中ERK1、ERK2及磷酸化的ERK(P ERK)蛋白的表达情况 ;应用免疫组织化学法分析其在细胞内的定位。结果 :肺癌组织中ERK1、ERK2及P ERK蛋白的表达水平明显高于癌旁正常组织 ,分别为癌旁组织的 1.5 ,1.8和 1.7倍 (P值均小于 0 .0 1) ;ERK1与ERK2蛋白表达水平呈线性正相关 (r=0 .6 4 ,P <0 .0 1)。免疫组织化学显示ERK蛋白主要位于细胞浆内。结论 :ERK蛋白的过表达可能在非小细胞肺癌的发生、发展过程中起着重要的作用。

Abstract:Objective: To investigate the expression and activation of extracellular signal-regulated kinases (ERK) in lung cancer and its clinical significance. Methods: Samples were obtained from 52 patients with nonsmall cell lung cancer. Western blot was used to measure the expression of ERK1、ERK2 and P-ERK protein.Immunohistochemistry was used for localization of ERK protein. Results:The levels of ERK1、ERK2 and P-ERK protein in nonsmall cell lung cancer tissues were increased,which were 1.5,1.8 and 1.7 times compared with those in adjacent normal tisses respectively,and they all have remarkable difference(P<0.01).ERK1 and ERK2 were observed to be a positive linear correlation(r=0.64,P<0.01). Immunohistochemistry showed that ERK protein were mainly located in cytoplasm. Conclusion: Overexpression of ERK may play an important role in the development of human nonsmall cell lung cancer.
