摘    要:目的 考察银离子抑菌的有效浓度及安全浓度范围,为抗菌制剂中银离子的使用浓度选择提供依据.方法 采用体外抑菌实验的方法检测不同浓度银离子的抑菌能力,采用体外细胞毒性噻唑蓝(MTT)法检测不同浓度银离子的细胞毒性.结果 银离子浓度低于12.5 ppm时无细胞毒性,但在25 ppm细胞存活率降为7%,细胞毒性在12.5~25 ppm之间急剧增大;银离子具有强效抑菌作用,在低浓度10 ppm可100%抑制金黄色葡萄球菌、大肠埃希菌、白色念珠菌、黑曲霉菌、铜绿假单胞菌的生长,且抑菌作用随浓度增加而增强.结论 银离子抗菌的最佳浓度为10 ppm.

Abstract:Objective To investigate the effective antibacterial and safe concentration range of silver ion,and provide the basis for concentration selection in disinfection products containing silver.Methods In vitro MTT assay was used to evaluate the cytotoxicity of Ag+ concentration gradient. In vitro inhibition assay was used to detect the antibacterial ability of silver ion concentration gradient.Results Below the concentration of 12.5 ppm,Ag+ was totally safe to L929 cells.But in 25 ppm,the cell viability rate falls to 7%,which indicated that cytotoxicity of Ag+ showed a rapid increase from 12.5 ppm to 25 ppm.Ag+ in 10 ppm inhibited Staphylococcus aureus,Escherichia coli,Candida albicans,Aspergillus niger and Pseudomonas aeruginosa growth completely,and the inhibitory effect increased with the increase of concentration.Conclusion 10 ppm was the optimal antimicrobial concentration of silver ion.
