内质网应激与炎症反应的研究进展

摘    要:细胞应激状态会严重扰乱内质网功能,引起未折叠或错误折叠蛋白质在内质网腔内异常积聚.内质网通过3条信号通路组成的未折叠蛋白反应来维持内质网稳态.本文综述了近年来内质网应激与炎症反应通过多种机制相互偶联,调节机体内稳态平衡或参与多种炎性疾病的研究进展.这些研究有助于阐明内质网应激介导炎症反应的精确机制,为研发以内质网应激-未折叠蛋白反应-炎症反应为靶点的药物提供理论依据.

Abstract:A variety of cellular stresses can severely perturb endoplasmic reticulum function,which cause pathological protein misfolding and aggregation within the ER.The unfolded protein response is a three-pronged signaling axis dedicated to preserving ER homeostasis.The paper reviewed the latest research progress on the complex interplay of ER stress,inflammation,and organismal homeostasis.Inflammation is both a cause and consequence of ER stress,and identifying additional signaling convergence points will guide the development of powerful therapeutics for inflammatory diseases and other UPR-mediated disorders with inflammatory components.
