摘    要:目的;探讨血清非免疫球蛋白成分10kD蛋白与重症肌无力的关系。方法(1)用SDS-PAGE分析健康人与MG患者的血清蛋白。(2)用抗体测血清中10kD蛋白水平。(3)观察10kD蛋白对培养的健康人及MG单核血细胞增殖的影响。结果(1)10kD与14kD扫描面积的比值存在明显差异,97%的健康人面积之比>1,而MG患者仅有30%。(2)10kD蛋白浓度在健康人及MG患者两组问不存在有意义的差别。(3)10kD蛋白100μg/ml可抑制健康人未激活的和PHA激活的单核血细胞增殖,但该蛋白和PHA对MG患者的细胞增殖无影响。结论(1)血清10kD与14kD蛋白扫描面积的比值可作为生化诊断MG的辅助参数。(2)10kD蛋白具有调节健康人单核血细胞增殖的生物活性。

Abstract:Aim: To explore characteristics of two non-immunoglobulin components with mass of about 10 kD and 14 kD proteins in healthy and patients with myasthenia gravis (MG). Methods: (1) The patterns of serum proteins taken from 35 healthy and 61 MG patients were analysed by SDS-PAGE. (2) Concentration of 10 kD protein in healthy and MG sera were determined by ELISA with polyclone antibody. (3) The biological activity of 10 kD protein was determined by observing its effect on proliferation of mononuclear blood cells. Results: (1) The scanning area ratio of 10 kD and 14 kD was found to be significant difference between the healthy and patients with MG. By taking the ratio > 1 for normal, the number of normat was about 97% in the healthy sera, but was only 30% in the MG sera. (2) No significant difference for level of 10 kD protein was found not only between healthy and MG sera, but also between seropositive and seronegative MG sera due to scattering concentration of the protein. (3) The proliferation of healthy mononuclear blood cells of both normal and stimulated with PHA was found to be obviously inhibition hy 10 kD protein (100 ug/ml), but no distinct effect on the MG. Conclusion: (1 ) The ratio of scanning area of 10 kD to 14 kD could be taken as an auxiliary index for biochemical diagnosis of MG. (2) The 10 kD protein possessed a function of regulating healthy cellular proliferation.
